Dysregulation of lipid metabolism in PLHC-1 and ZFL cells exposed to tributyltin an all-trans retinoic acid

چکیده
There is increasing awareness that exposure to endocrine disrupters interferes with lipid homeostasis in vertebrates, including fish. Many of these compounds exert their action by binding nuclear receptors, such as peroxisome proliferator-activated receptors and retinoid X receptor. This work investigates the use fish liver cells (PLHC-1 ZFL cells) for screening metabolic aquatic environment assessing changes cell’s lipidome after model compounds, tributyltin chloride all-trans retinoic acid. Lipid extracts, analyzed FIA-ESI (+/-) Orbitrap, evidenced intracellular accumulation triglycerides diglycerides both cell models 100 200 nM 24 h. Exposure 1 μM acid led a significant PLHC-1 cells, while few were accumulated cells. Retinoic (cyp26b1, cyp3a65, lrata) metabolism (fasn, scd, elovl6) related genes up-regulated acid, only down-regulated expression dgat1a. The two show sensitivity responses comparable those previously reported mammalian These results support alternative detection contaminants act environment.

All-trans-retinoic acid: measurement
A quantitative determination of physiological levels of all-trans-retinoic acid (vitamin A acid) in human serum has been developed. A double-phase extraction of 3.5 ml of serum followed by a specific and sensitive high performance liquid chromatography procedure allowed measurement of levels down to 1 ng/ml. Serum concentrations in 37 fasting volunteers ranged from 2.7 to 4.2 ng/ml and fitted a...
متن کاملProphylactic effect of all-trans retinoic acid in an amyloid-beta rat model of Alzheimer\'s disease
Introduction: Retinoid signaling has been argued to have favorable effects on Alzheimer's disease (AD). We studied the role of chronic intracerebroventricular (ICV) injection of all-trans retinoic acid (ATRA) on the amyloid-beta (Aβ) model of AD. Methods: Adult male rats weighing 260-330 g were divided into 12 groups of 8 each. Six groups of rats received ATRA (3nM, 30nM, 3μM, 0.3mM, 30...

These effects have become a therapeutic objective in human cancers. In addition to these functions, ...
متن کاملRetinol inhibits the growth of all-trans-retinoic acid-sensitive and all-trans-retinoic acid-resistant colon cancer cells through a retinoic acid receptor-independent mechanism.
Retinol (vitamin A) is thought to exert its effects through the actions of its metabolite, all-trans-retinoic acid (ATRA), on gene transcription mediated by retinoic acid receptors (RAR) and retinoic acid response elements (RARE). However, retinoic acid resistance limits the chemotherapeutic potential of ATRA. We examined the ability of retinol to inhibit the growth of ATRA-sensitive (HCT-15) a...
